Research question

How does the severity of cardiovascular-kidney-metabolic (CKM) syndrome relate to dementia risk, cognitive performance, and brain structure?

Study

The study analysed 387,340 UK Biobank participants across five CKM stages. Cox and linear regression models examined incident dementia, cognitive decline, and neuroimaging outcomes, followed by mediation analysis of structural brain measures.

Key finding

More advanced CKM stages were associated with higher dementia risk, cognitive decline, lower total brain, gray-matter, and hippocampal volumes, and greater white-matter hyperintensity volume. Structural brain changes partially mediated the CKM-dementia association.
